美國PolyLC PolyETHYL A色譜柱 HIC分離蛋白的方法是基于蛋白質(zhì)的疏水特點(diǎn)，比如RPC。但是，HIC使用完q含水的緩沖器,保留住了蛋白質(zhì)的三級結(jié)構(gòu)和生物活性。這種分離性通常優(yōu)于RPC方法分離蛋白質(zhì)和多肽，能夠極大的保留蛋白質(zhì)的二級結(jié)構(gòu)和三級結(jié)構(gòu)。通常情況下,樣品使用硫酸鹽或磷酸鹽等鹽濃度降低梯度洗脫，通過增加蛋白質(zhì)表面疏水性將其洗脫。 PolyLC美國聚合物有限公司專業(yè)化生產(chǎn)用于蛋白質(zhì)，多肽和其他生物大分子分析的反相高效液相色譜固定相的替代品。 HIC分離蛋白的方法是基于蛋白質(zhì)的疏水特點(diǎn)，比如RPC。但是，HIC使用完q含水的緩沖器,保留住了蛋白質(zhì)的三級結(jié)構(gòu)和生物活性。這種分離性通常優(yōu)于RPC方法分離蛋白質(zhì)和多肽，能夠極大的保留蛋白質(zhì)的二級結(jié)構(gòu)和三級結(jié)構(gòu)。通常情況下,樣品使用硫酸鹽或磷酸鹽等鹽濃度降低梯度洗脫，通過增加蛋白質(zhì)表面疏水性將其洗脫。表面活性劑(例如丙磺酸；辛基糖苷)如果必要的話可以被添加到流動相的。PolyPROPYL A™，PolyETHYL A™和PolyMETHYL A™的相對疏水值分別是100, 60和15。HIC比其他方法有更大的敏感性，尤其是涉及到非極性基團(tuán)。HIC適用于: l 多維蛋白純化（建議順序:離子交換-鹽梯度洗滌分離-HIC)。 l 多肽的純化（比如毒液、糖肽類） l 表面抗體 l 質(zhì)量控制分析蛋白質(zhì)的單一殘基的極性或者突變體的修飾位點(diǎn)。 大于20KDa的蛋白質(zhì)建議使用1000 -或1500-A的孔。其能力可以與離子交換相媲美。除非該蛋白是g認(rèn)的顯著疏水性，建議使用PolyPROPYL A™。
